Trout testis cells. 3. Acetylation of histones in different cell types from developing trout testis.

چکیده
Acetylation of histones has been observed in both the spermatocytes and spermatids from developing trout testis. In the former cell type, both histone synthesis and extensive acetylation occur, whereas in the spermatids there is histone acetylation, but very little synthesis. The half-life of the histone acetyl groups is approximately 23 hours in the prespermatid cells. It is suggested that the acetylation of histones in spermatids may serve a different function from the corresponding modification observed in the earlier testis cells.

Multiacetylated forms of H4 are found in a putative transcriptionally competent chromatin fraction from trout testis
We have examined the distribution of acetylated histones derived from various trout testis chromatin fractions of different composition. Our results indicate that a chromatin fraction, preferentially solubilized by micrococcal nuclease, containing the bulk of the HMG proteins and similar to a fraction released from intact trout nuclei and previously shown to be enriched in transcribed DNA seque...
متن کاملThe subunit structure of chromatin: characteristics of nucleohistone and nucleoprotamine from developing trout testis.
Recent work has demonstrated the existence of a subunit structure in chromatin. Hewish and Burgoyne have reported [I] that a large proportion of DNA in isolated rat liver nuclei is digested by an endogenous Ca++ and Mg+’ dependent endonuclease into fragments which are integral multiples of a unit length. Louie has found [2] that DNA from mouse liver nuclei or from the nucleoprotein complex of p...
متن کاملSites of in vivo histone methylation in developing trout testis.
Specific lysyl residues of trout testis histones H3 and H4 are methylated partially during rainbow trout spermatogenesis. Histones H1, H2A, H2B, and protamine are not methylated. The single site (lysine 20) in histone H4 and the two major sites (lysines 9 and 27) in histone H3 are homologous to those determined for other organisms, but an additional minor site (lysine 4) occurs in histone H3. A...
متن کاملModification of histones during spermiogenesis in trout: a molecular mechanism for altering histone binding to DNA.
At a late stage of spermatogenesis in rainbow-trout testis, the entire complement of histones is replaced by newly synthesized protamine and histones are extensively phosphorylated and acetylated. Tryptic digestion of purified histones labeled by incubation of testicular cells with [(32)P]phosphate shows that phosphorylation occurs at a small number of seryl residues. Histone I (lysine-rich) is...
متن کاملModification of Histones during Spermatogenesis in Trout
Trout testis cells were incubated with 13H]arginine and lysine and inorganic [32P]phosphate. Histones were extracted with acid and the major fractions (I, IIbl, IIb2, III monomer, and IV) were resolved by chromatography on BioGel P-10. The profile of 32P incorporation was time dependent. When the phosphorylated histones in each of the major fractions were resolved from unphosphorylated species ...
